A mediocre first half performance when the Spireites went two goals down was more than eclipsed after John Sheridan made nine changes at the break which led to the game being turned on its head with three goals in the opening 25 minutes of the second half.
Summer signings Dwayne Mattis and Danny Whitaker made their Chesterfield mark with powerful efforts that proved to be too much for the Gladiators.
Mattis had a late chance to take the match ball home after Jack Lester was upended in the penalty box, but the midfielder's effort was matched by the Matlock number one.

Mark Allott collected the Banner Jones Cup for the Spireites as they picked up their first piece of silverware of the season.
Manager John Sheridan decided to stick with trialist Shane Redmond in between the posts as Tommy Lee remained on the bench for the second consecutive match. Amongst the starting eleven were a further two trialists, Sean Timmons who featured against Alfreton Town at the Impact Arena at the weekend and right back Jack Hunt who is currently on trial from Yorkshire outfit Huddersfield Town.
Summer signing Simon Ford and youngster Dan Gray completed the back four with an even more youthful midfield. Long serving midfielder Derek Niven teamed up with three first year pro's Craig Clay, Jimmy Adcock and Tendayi Darikwa, while Jordan Bowery and Scott Boden completed the team upfront.

Just 10 minutes after the kick off and the hosts took the lead through ex-Spireite Ben Algar. A wicked deflection from Algar beat Redmond to gift the Gladiators the early lead.
The Gladiators found the back of the net again on the stroke of half time as a bullet header from James Knowles proved too much for the Spireites to handle as John Sheridan and his side went in at the break two down.
Half Time: Matlock Town 2 Chesterfield 0
As expected, Sheridan made a number of changes at the half time interval introducing nine players into the fold. This introduction had a positive impact on the match as Danny Whitaker struck the post just minutes after replacing Darikwa on the left wing.
The Spireites continued their positive approach to the second half as substitute Dwayne Mattis pulled a goal back for Chesterfield with a fierce drive from the edge of the box just five minutes after the re-start.
Mattis found the back of the net again to level the game up with another powerful strike from the edge of the Matlock box.
Danny Whitaker added his name to the score line on 59 minutes with a rasping shot from outside the box to turn the match around for the Spireites.
Both Chesterfield and Mattis could have extended the scoreline further after veteran Jack Lester was upended to award the Spireites a penalty, but unfortunately Mattis couldn't make the match ball his as he was denied by the Matlock number one.
