Dave Bentley's young troops will be in action at Saltergate on Tuesday night in the Midland Youth Cup second round.
The Spireites were taken the distance by a tough Kidderminster opposition in the previous round before Chesterfield killed the game off in extra time through Tendayi Darikwa's half volley.
Chesterfield currently still remain in two cup competitions with the only exit being the FA Youth Cup defeat against Northampton.
Opposition on Tuesday night comes in the form of Nuneaton Town, who made it to the second round with a 3-2 victory over Stratford Town in the earlier round.
Bentley's side have recently finished their league campaign on a high until after the New Year, with an away win over Hartlepool United at the weekend.
Kick off on Tuesday night is 7.30pm and admission to the main stand is free.
